Course content
Lectures 1. Blood and bone marrow, function of cellular blood elements in peripheral blood, nomenclature. Physiological values of blood elements. Physiology of hematopoiesis, proliferation, differentiation and maturation of individual hematopoietic lineages. 2. Development, function and destruction of erythrocytes (hemoglobin synthesis and function, iron metabolism, breakdown products of Hb during intravascular and extravascular hemolysis). 3. Development and function of leukocytes, morphological differentiation of leukocytes. 4. Platelet function, primary hemostasis. 5. Blood clotting and its stages. 6. Mechanisms ensuring hemostasis, coagulation and fibrinolytic system, system of natural inhibitors. 7. Blood count and hemocoagulation screening tests. 8. Blood particle analyzers and coagulometers, screening and special coagulation tests, calibration and validation of hemocoagulation methods. 9. Laboratory diagnosis of blood diseases and disorders of hemostasis, differential diagnosis. 10. Pathophysiology of blood diseases and disorders of hemostasis. 11. Tumor disorders of hematopoietic cells - leukemia, myelodysplastic syndrome, myeloproliferative disease, lymphoproliferative disease. A Lab exercise 1. Principles of proper laboratory work. 2. Examination of the blood picture on an automatic hematological analyzer. 3. Examination of peripheral blood smears microscopically, determination of the number of blood elements in the counting chamber. 4. Microscopic examination of bone marrow smears. 5. Investigation of hemolysis including osmotic resistance of erythrocytes. 6. Examination of platelet aggregation on an aggregometer. 7. Examination of basic coagulation tests - prothrombin time and activated partial thromboplastin time on a coagulometer. 8. Determination of factors of the plasma system of blood coagulation. 9. Determination of inhibitors of the plasma system of blood coagulation. 10. Hemoglobin electrophoresis, determination of the von Willebrand factor by the LIA method, evaluation of the outputs of hematology analyzers, including interferences. 11. Cytochemical staining methods.

Knowledge
explains the physiology of hematopoiesis and blood coagulation and, on this basis, clarify the essence of malignant and non-malignant blood diseases and blood coagulation disorders
explains the principles of laboratory diagnosis of blood diseases and coagulation disorders and interprets the possibilities and limits of instrumental blood picture analysis and coagulometers
Skills
performs an instrumental analysis of the blood picture
makes a smear of peripheral blood and bone marrow and evaluates it microscopically
performs instrumental screening coagulation tests and evaluates them
controls basic operation and daily operation of analyzers and coagulometers
calibrates the instruments and sets the daily norm
differentiates physiological from pathological results and performs basic differential diagnosis of blood diseases
