Course content
A Lab exercise 1. Introduction to work in a biochemical laboratory. OSH, PO 2. Reaction of basic functional groups of organic compounds. 3. Water and pH. Buffers, buffering capacity. pH measurement. 4. Properties of amino acids - determination, concentration, TLC 5. Determination of protein concentration - Biuret reaction, Lowry's method. Properties of protein solutions - isoelectric point, denaturation. 6. Separation of low-molecular and high-molecular substances - dialysis, gel chromatography. 7. Measurement of enzyme activity - effect of temperature, activity, specific activity. 8. Determination of basic kinetic parameters of enzyme reactions - Michaelis constant, maximum reaction speed. 9. Inhibition of enzyme activity - determination of inhibitory concentration IC50, influence of inorganic ions. 10. Myoglobin and hemoglobin - properties, determination, spectrophotometry. 11. Cholesterol and bile pigments. 12. Properties and determination of sugars, optical properties. 13. Properties of lipids. Biochemical examination of hyperlipoproteinemia. Lipoproteins (clinic). 14. Properties of nucleic acids. Isolation of DNA. Determination of DNA concentration and purity. 15. Credit test.
